Brief Description of Project:
Sugammadex (Org 25969) is a novel cyclodextrin compound which binds the non-depolarizing muscle relaxant rocuronium to reverse residual neuromuscular blockade at the end of surgery. Standard neuromuscular reversal drugs, namely neostigmine and edrophonium, require co-administration of an anticholinergic drugs (namely, glycopyrrolate and atropine, respectively) to minimize their cardiovascular side effects (e.g., profound vagally-mediated bradycardia). In contrast, sugammadex has no known cardiovascular effects.
The current study is designed to evaluate the time course of reversal of a rocuronium-induced neuromuscular blockade and the changes in heart rate and blood pressure after a bolus dose of sugammadex. Specifically, we will determine the time-course of recovery to a T4/T1 ratio to 0.9 when sugammadex, 4.0 mg/kg IV is administered after rocuronium as an alternative to neostigmine/glycopyrrolate in a wide range of surgical procedures and anesthetic regimens to mimic routine clinical practice.
