The focus of research in my laboratory
has been to understand how the male hormones, testosterone and
5-alpha-dihydrotestosterone exert their effects during embryogenesis, in
the adult, and in disease states. These interests are focused into the following
broad areas:
1) Androgen resistance: We have defined
a large number of mutations in the androgen receptor (AR) gene that cause
androgen resistance. It is clear that most are caused by amino acid substitutions
in the hormone-binding domain of the receptor. Current work is focused on
understanding how these substitutions interfere with AR function at the molecular
level.
2) Genetic control of androgen receptor
expression: AR expression varies from tissue to tissue and at different times
during development. The basis for those controls is being defined using cell
culture and transgenic models.
3) The androgen receptor in human prostate
cancer: The clearcut relationship between prognosis and estrogen receptor
expression in breast cancer does not hold true in all patient groups for
AR expression and prostatic cancer. It is clear, however, that AR expression
does vary in selected prostate cancers. We are attempting to define the
implications of these changes and the mechanisms by which prostate cancers
progress to a state in which androgens are no longer required for growth.
4) Androgen metabolism and its effects
on androgen action: Testosterone is the principal circulating androgen in
mammals. It can undergo 5-alpha-reduction to for 5-alpha-dihydrotestosterone,
a more potent androgen, or aromatization to form estradiol, a potent estrogen.
Our work in this area is focused on a) characterizing genes that are
differentially regulated by testosterone and dihydrotestosterone and b) defining
the mechanisms controlling aromatase gene expression in rat gonadal cells.

Although my practice includes a range of patients, I have a particular interest in the diagnosis and treatment of patients with disorders of the pituitary and hypothalamus, such as those caused by pituitary adenomas. I work closely with the faculty of the Neurosurgery Department at UT Southwestern to organize preoperative evaluations and post operative care.
