PROCEDURE FOR DEVELOPING ELECTRON MICROSCOPY NEGATIVES (KODAK SO-163 FILM)

1. Developer -D-19 with continuous agitation for 4 minutes. (1:2 dilution developer:water; add 1000 ml developer to 2000 ml water- see procedure manual for details on how to make all photographic solutions).

2. Rinse in running water for 1.5 minutes with agitation. This rinse step is very important because it will help avoid mottle, unevenness, and stain due to excessive retention of developer in the emulsion.

3. Fix in Kodak Fixer for 10 minutes with frequent agitation.

4. Wash for 30 minutes in running water, sufficient to give one complete change of water every 5 minutes. To minimize drying marks, rinse 1 minute in Kodak Photo-Flo solution after washing. Or use Rapid Wash to reduce washing time and conserve water. Remove excess fixer by rinsing in water for 30 seconds; then bathe the film in Kodak Hypo clearing agent for 2 minutes with moderate agitation. Wash for 5 minutes in a water flow sufficient to give at least one complete change of water. Rinse for 1 minute in Kodak Photo-Flo solution to promote uniform drying.

5. Dry in a dust-free area or in a dryer with normal relative. To accelerate drying, use a heated cabinet or a current of warm air. Avoid excessive heat; a temperature of 100oF(38oC) is recommended.

6. D-19 stock is good for about 2 months. Diluted 1:2 D-19 is good for about 1 month in a gallon tank. It will process about 100 negatives. Fixer stock is good for about 2 months. In a gallon tank it is good for about 1 month. It will process about 150 negatives. Hypo clearing agent stock is good for about 3 months while the working solution is good about 24 hours. It will process about 75-90 negatives. Photo-Flo is good only for 24 hours.
